Simultaneous Determination of Glucose and Maltose in Sikhe using Oxygen Electrode with Dual Cathode System

Abstract

|
A dual cathode system was prepared with an oxygen electrode having one anode and dual cathode. Glucose oxidase for one cathode and amyloglucosidase and glucose oxidase for the other cathode were immobilized to determine glucose and maltose simultaneously. The components were determined by enzyme sensor was based on the maximum slope of oxygen consumption from the enzyme reaction. The response time for analysis was 1 min. The optimum condition for glucose and maltose sensor was found to be 0.1 M sodium acetate buffer, pH 5.0¡-5.5 at 35¡É. Various interferences such as sugar were investigated. The error in less than 5.5% was found in determination of maltose. The coexisting glucose in sample was compensated by the modified equation. This system was used for simultaneous determination of glucose and maltose in commercial sikhe and traditional one, which was confirmed by conventional methods. There was a good correlation between this method and spectrophotometry.
